摘要 [Objectives] To elucidate the mechanisms by which ginsenoside Rh2 inhibits the proliferation of breast cancer cells. [Methods] The effects of ginsenoside Rh2 on bone marrow-derived dendritic cells (BMDCs) in breast cancer-bearing mice were assessed using network pharmacology analysis, cytological assays, and protein histology techniques. The regulatory mechanisms of ginsenoside Rh2 on BMDCs were further explored via flow cytometry, enzyme-linked immunosorbent assay (ELISA), Western blot (WB), and reverse transcription polymerase chain reaction (RT-PCR). [Results] Network pharmacology analysis identified ginsenoside Rh2 as a potential therapeutic agent that facilitates dendritic cell (DC) maturation, with phosphoinositide 3-kinase regulatory subunit 1 (PIK3R1) serving as a crucial mediator. Experimental results confirmed that ginsenoside Rh2 enhanced BMDC maturation in breast cancer models via the PIK3R1-PDK1 signaling pathway. This enhancement coincided with decreased mRNA and protein expression levels of PIK3R1 and PDK1 in mouse DCs. [Conclusions] This study elucidates the potential molecular mechanisms of ginsenoside Rh2 through network pharmacology, which are subsequently validated by comprehensive laboratory experiments. Our findings indicate that ginsenoside Rh2 enhances tumor immunity and exerts anti-tumor effects by promoting DC maturation. Compared to conventional chemotherapy, ginsenoside Rh2 significantly reduces off-target toxicity while improving anti-tumor efficacy. In summary, this research provides novel insights and a theoretical foundation for the selection and development of breast cancer treatment regimens.
